چکیده

The increased demand in vegetable oil for food purposes and high-protein feed livestock poultry encourages producers to expand the production of various crops, while occupying rather cold agroclimatic zones. Improved frost resistance cultivated crops would significantly increase yield range rape cultivation a number climate regions. Nine transgenic lines winter containing genes encoding proteins with shock domain (CspA и EsCSDP3) were obtained as result Agrobacterium transformation. In total, 260 explants involved transformation using pBI121-CSPA-plant, efficiency 2.3%; among 750 pBI-EsCSDP3 construction, was 0.4%. As studies, it shown that expression new gene Escsdp3 from plant Eutrema salsugineum able plants effectively cspa E. coli, which is classically used this purpose. analysis T1 generation revealed four resistant (three cspA-plant one line gene). transfer into led significant plants. Two rapeseed freezing (with Non-hardened remained viable after 24 h exposure negative temperatures up −5 °C, passed through hardening stage survived at −16 °C.

Evaluation of Cold Tolerance in Rapeseed (Brassica napus L.) Using Stress Tolerance Indices

In order to study the response of 120 genotypes of rapeseed (including 100 hybrids and 20 parents) in terms of cold stress tolerance, an experiment was conducted in α-lattice design with two replications under normal (September) and late sowing (October) conditions during two years. Combined analysis of variance showed that genotypes were significantly different at 1% and there was a significan...

High DNA melting activity of extremophyte Eutrema salsugineum cold shock domain proteins EsCSDP1 and EsCSDP3

Plant cold shock domain proteins (CSDP) participate in maintenance of plant stress tolerance and in regulating their development. In the present paper we show that two out of three extremophyte plant Eutrema salsugineum proteins EsCSDP1-3, namely EsCSDP1 and EsCSDP3, possess high DNA-melting activity. DNA-melting activity of proteins was evaluated using molecular beacon assay in two ways: by me...

Adenine Nucleotide Changes during Cold Acclimation of Winter Rape Plants.

During the first stage of hardening of winter rape plants (Brassica napus L. var. oleifera L., cv. Górczański), marked increase of ATP content in leaves was observed. Lowering the temperature from 5 to 0 C (the second stage of hardening) had no further effect on ATP content. In roots, not capable of hardening, pronounced decrease of ATP content was noted after prolonged exposure to cold. It was...

Lipid remodelling: Unravelling the response to cold stress in Arabidopsis and its extremophile relative Eutrema salsugineum

Environmental constraints limit the geographic distribution of many economically important crops. Cold stress is an important abiotic stress that affects plant growth and development, resulting in loss of vigour and surface lesions. These symptoms are caused by, among other metabolic processes, the altered physical and chemical composition of cell membranes. As a major component of cell membran...
